Based on the knowledge that hexsosaminyl linkage is much more\nresistant to acid cleavage than neutral sugar linkages (include N-acetylhaxsosaminyl), this linkage therefore could be distinguished from other glycosyl bonds. Hydrazinolysis is used to selectively cleave amide bonds in protein and has been applied to the synthesis of radioactive derivative by\nre-acetylation of de-N-acetyl glycosphingolipids.\nIn this experiment, we examined whether controlled hydrazinolysis could be used for de-N-acetylation of complex glycosphingolipids. Ceramide tri-, tetra, and pentasaccharides are readily converted to their de-N-acetyl glycolipid deriavatives as shown by TLC and MALDI-TOFMS analysis. And we confirmed by GC analysis that hexosaminyl linkage are resistant to acid cleavage.\nFinally, we have applied this technique to an example of complex glycosphingolipid,\nnamely ceramide nonasaccharide, and have detected its complete sugar linkager which agree with its previously determined branching structure.
